The predecessor of the 250th Air Defense Brigade was the Yugoslav 250th Anti-Aircraft Missile Regiment. In 1962, the Yugoslav Air Force formed the 250th Air Defense Regiment, which was specifically responsible for the air defense of the capital Belgrade, with 4 missile battalions and 1 technical support battalion, equipped with Soviet-made S-75 air defense missiles. In 1980, the 250th Air Defense Regiment was expanded into a brigade-level combat unit and replaced with S-125 anti-aircraft missiles.

In 1999, with NATO's indiscriminate bombing of the Federal Republic of Yugoslavia, the 250th Air Defense Brigade stationed in Belgrade naturally became a "key target of care", according to incomplete statistics, NATO troops carried out more than 100 targeted attacks on the 250 Air Defense Brigade.
