The "black hand behind the scenes" of the new crown virus has been found
The research team of Professor Yu Wenqiang, a researcher at the Institute of Biomedical Sciences of Fudan University and the Shanghai Public Health Clinical Center, and the research team of Professor Lu Hongzhou, president of the Third People's Hospital of Shenzhen, have found that the "human sequence" in the new coronavirus is the key factor in the pathogenesis after nearly two years of intensive research. These common "codes" of the new crown virus and humans can not only activate the expression of inflammation-related genes and induce cytokine storms, but also promote the accumulation of hyaluronic acid in the body, resulting in the production of clinical symptoms of new crown pneumonia such as pulmonary grinding glass lesions. The study also found that oral administration of oxymethotodouin, which inhibits hyaluronic acid, is expected to become a new means of treatment for the new crown. Recently, the latest issue of The Lancet sub-journal "eBioMedicine" ("Electronic Biomedicine") published this research results.
Yu Wenqiang introduced that the new crown virus belongs to the RNA virus, the most important feature is constantly mutated, and the drug research and development for the new crown must outperform the continuous mutation of the new crown. The study included 137 patients with COVID-19, and through sequence comparison analysis, it was found that 5 segments of the new coronavirus genome had exactly the same sequence as humans. Genetic sequences are the "code" for all life forms. The study confirmed that the new crown virus has a common "code" with humans. The research team named this genetic sequence of the new coronavirus "humanoid sequence."
What exactly does the "sequence of people" play? Further research found that the "humanoid sequence" is located in a seat of a gene called an enhancer in the human genome, and the enhancer is thought to be a switch in human gene expression. Interestingly, these "human sequences" are "conservative" in 159258 coronavirus genomes, meaning that the "code" common to humans in these viruses does not change, regardless of how the viruses mutate. This suggests that the "human sequence" is the "behind-the-scenes" of all the new coronavirus diseases.
Based on previous research work, the research team found that the "human sequence" in the virus can form potential microRNA precursors that can activate inflammation-related gene expression in different cells. This suggests that the "humanoid sequence" is the key to inducing a storm of COVID-19 cytokines. At the same time, the study found that the "human sequence" can also activate hyaluronine synthase HAS2 in vivo and induce the accumulation of hyaluronic acid, while the clinical symptoms and sequelae of new crown pneumonia patients (including grinding glass lesions and thrombotic diseases in the lungs) are mostly related to the accumulation of hyaluronic acid. Surprisingly, the team's treatment of hyaluronic acid alone was able to cause pulmonary terrazzo lesions in mice, suggesting that hyaluronic acid may be related to specific lung lesions in patients with new coronary pneumonia.
Yu Wenqiang said that hyaluronic acid, also known as hyaluronic acid, is the "darling" of today's beauty industry, but it is the "culprit" of the new crown. Studies suggest that hyaluronic acid is an important marker for predicting the progression of the disease of patients with new coronary pneumonia, and the detection of plasma hyaluronic acid content is expected to become an important indicator for screening asymptomatic infected people and judging whether patients need to be admitted to hospital.
The research team believes that hyaluronic acid can be used as a target to develop new crown treatment drugs. Along these lines, the researchers found oxymethocoumarin, an oral inhibitor of hyaluronic acid. The drug is clinically used in the treatment of cholecystitis and other treatments, with low pharmacological toxicity and high safety, which can significantly inhibit the hyaluronic acid accumulation mediated by the "human sequence" of the new coronavirus. Initial clinical trials have confirmed that oral oxymethodalamarin improves clinical symptoms such as reduced lymphocyte count and lung lesions in patients with COVID-19. The study explores a new approach to COVID-19 treatment.
