In the study, a detailed pharmacokinetic study was carried out on the bioavailability of vitamin D₃ oral nanospray. The pharmacokinetic properties of administration by oral spray and conventional oral administration were compared using New Zealand white rabbits as animal models. The results of the study showed that the highest concentration (Cmax) of 25-OH-VitD3 in the plasma of rabbits administered with the spray form was 1.7 times higher than that of oral administration. The highest concentration (Cmax) of 25-OH-VitD3 in the plasma of rabbits administered with oral spray was significantly higher than that administered orally, reaching 139.93 ng/mL, while the Cmax of oral administration was 55.88 ng/mL. In addition, the peak time to arrival (Tmax) of oral spray administration was 26 hours, which was shorter than the 40-hour period of oral administration. These data suggest that vitamin D₃ oral sprays have a clear advantage in bioavailability, increasing drug concentrations in the body faster and more effectively than traditional delivery methods.
