The efficiency of gene editing and detection of watermelon was improved

Recently, the watermelon genetics, breeding and cultivation innovation team of Zhengzhou Institute of Pomology, Chinese Academy of Agricultural Sciences has developed a rapid, stable and efficient method to detect the gene editing efficiency of the guide target in the genus Watermelon using an Agrobacterium rhizoides-mediated transformation system, and successfully verified the editing efficiency of the target in the stable genetic transformation system of watermelon. The findings were published in the Horticultural Plant Journal.

Gene editing is a technology that specifically modifies the target gene, and is widely used in biological breeding, gene function verification and other research fields. Guide RNA is a key component of the gene editing system, and its editing efficiency significantly affects the workload of genetic transformation.

In order to quickly and efficiently detect the editing efficiency of guide RNA targets, a watermelon protein kinase gene was used as the target gene construction vector, and Agrobacterium rhizogenes induced watermelon to rapidly produce a large number of hairy roots edited by guide RNA targets, and the editing efficiency of guide RNA targets was successfully evaluated in Watermelon. In stably transformed watermelon seedlings, 100% editing of the guide RNA target occurred. The development of this method can significantly reduce the workload of watermelon gene editing and accelerate the progress of watermelon functional gene validation.
