In 1927, he was appointed chief of staff of the 74th Regiment of the 25th Division of the 4th Army of the National Revolutionary Army. In August of the same year, he led the heavy machine gun company of the regiment to participate in the Nanchang Uprising. After the uprising, he served as the commander of the 74th Regiment and went south to Guangdong with his troops. At the end of October, the remnants of the Nanchang rebel army were reorganized into a column, with Wang Erzhuo as chief of staff, and together with Zhu De, Chen Yi, and others, he moved to Fujian, Guangdong, Gansu, and Xiangbian, and persisted in armed struggle. In January 1928, he participated in leading the Shonan Uprising and served as chief of staff of the 1st Division of the Workers' and Peasants' Revolutionary Army.

In April 1928, after Zhu De and Mao Zedong's jinggangshan division met, Wang Erzhuo was appointed chief of staff of the 4th Army of the Chinese Workers' and Peasants' Red Army and commander of the 28th Regiment. He assisted Mao Zedong and Zhu De in commanding the Red 4th Army to win victories in the battles of Wudoujiang, Caoshiao, and Longyuankou, and smashed the second, third, and fourth large-scale "advances" launched by the Kuomintang army. Wang Erzhu led the 28th Regiment to fight bravely, became a member of the Jinggangshan generals, and made major contributions to the defense and development of the Jinggangshan revolutionary base area. On August 25 of the same year, when pursuing the traitors at the Chongyi Sishun Ruins in Jiangxi, he died heroically at the age of 25.

At the memorial service for Wang Erzhuo, Mao Zedong and Zhu De spoke highly of his contributions to the revolution. Hanging in the venue was a link drafted by Mao Zedong and written by Chen Yi: "One cry Erzhuo, two cry Erzhuo, Erzhu is now finished!" Who will bear the burden of staying? Born as a class, died as a class, how about after class? Get the victory of the beginning of the holiday! ”

In 2009, Wang Erzhuo was elected as one of the "100 Heroic And Exemplary Figures Who Made Outstanding Contributions to the Founding of New China".

"Wang Erzhuo is a famous heroic martyr who walked out of the old Shimen District and is an excellent commander of the Chinese Workers' and Peasants' Red Army. In his short but brilliant life, he vividly explained the unswerving pursuit of faith, the heroic and tenacious spirit of struggle, and the revolutionary pride of not being afraid of sacrifice. Liu Fanbiao, deputy director of the Shimen County Party History Research Office, said.

Shimen County is located on the border of Xiang'e, at the northeast end of the Wuling Mountains, with dangerous terrain and high altitude, and was once included in the counties of the Wuling Mountains concentrated in contiguous areas of extreme poverty. Today, Shimen County has lifted itself out of poverty, and the former "poor mountain nest" has undergone earth-shaking changes. At the end of 2020, with the completion and trial operation of the Shimen to Cili section of the Anci Expressway, Shimen County ended the history of no highway and forged ahead on the avenue of high-quality development.
