ASO announced the teams participating in the Women's Paris-Roubaix Classics in France on April 16 and the Paris-Roubaix Classics on April 17. In addition to the teams that were automatically invited, 7 women's teams and 4 men's teams received wild card invitations for the event.
In the inaugural race last October, Lizzie Deignan from the Trirk-SagaLando women's team won the first race after flying 82 kilometres solo to lift the stone trophy. The stone trophy at last year's men's 118th Paris-Roubaix Classic was won by Sonny Colbrelli from Bahrain's Victory Team.
Trek-Segafredo will be one of the 14 women's World Tour teams participating, along with Canyon-SRAM Racing, FDJ Women's Team (FDJ Nouvelle-Aquitaine Futuroscope), LIV Racing Xstra, and Movistar Team Women), Team BikeExchange-Jayco, DSM Women's Team (DSM), SD Worx Women 's Team (SD Worx), EMIRATES Women's Team (UAE Team ADQ), as well as the new top teams Jumbo-Visma, Uno-X, Roland.) The three highest-ranked intercontinental pro teams were also automatically invited, including the Ceratizit-WNT, the Parkhotel Valkenburg and the Valcar-Travel & Service.
Seven intercontinental professional teams were invited to wild cards, including four French Arkéa Pro Cycling Team, Cofidis Women Team, Stade Rochelais Charente-Maritime and St Michel-Auber 93, as well as Britain's Le Col Wahoo Team. The NXTG women's team in the Netherlands (NXTG by Experza) and Belgium's Plantur–Pura women's team.

The men's race will feature 18 World Tour teams, including defending champion team Bahrain Victory, as well as AG2R Citroen, Astana-Qazaqstan, Bora-Hansgrohe, Cofidis, QuickStep-AlphaVinyl, EF Education-EasyPost, and EF Education-EasyPost. Vincenzo Nibali, who represents Astana, plans to compete in all five classics in 2022, including Paris-Roubaix.
